Rutin is a flavonoid compound which are found in watermelon and peel of citrus plants. Rutin had many pharmacological activities, one of them as anti-rheumatoid arthritis. Rutin showed a low absorption when administered orally because of it had a low permeation and can be degradation in the gastrointestinal tract, so the alternative administered of rutin by transdermal route. Transdermal is a route of drug delivery that support drug through skin into the systemic circulation. This route could prevent degradation of rutin in gastrointestinal track. Transfersom is one of carrier for transdermal route that can improve the effectiveness of drug delivery. This study aims to evaluated in-vitro penetration test and anti-rheumatoid arthritis activity from gel which contained rutin loaded with transfersom and gel contained rutin without loaded transfersome. In-vitro penetration test showed that the penetration of rutin loaded with transfersom gel was 14.33%, while for non transfersom gel was 9.51%. Anti Rheumatoid Arthrithis activity was observed in 28 days with calculated the percent inhibition of edema volume, the percentage inhibition of transfersom gel was 57.82% while non transfersom gel was 45.63%.
