This study was conducted to isolate and test the antibacterial activity of bacterial isolate X2 associated with Xestospongia testudinaria sponge from Pasir Putih Beach Situbondo. Bacterial isolation was carried out by pour plate method then bacteria colony obtained was purified by streak plate method. Isolate bacteria X2 produced 12 bacterial isolates which was tested for antibacterial activity against test bacterias of Klebsiella pneumonia, Pseudomonas aeruginosa and Salmonella typhii using overlay method. The results demonstrated that obtained isolate X2-10 was active against P. aeruginosa ATCC27853 and MDR using agar diffusion method. Characterization of isolate X2-10 was done by Gram staining test, biochemical test and identification with 16s-rRNA. The results showed that bacterial isolates X2-10 associated with X. testudinaria sponge displayed an antimicrobial activity against P. aeruginosa ATCC27853 and MDR. Based on the characterization result, the bacterial isolate X2-10 was B. licheniformis.
