Papaya seed contains vitamin E and flavonoid that have antioxidant activity. This study was aimed to determine the antioxidant effect of ethanol extract of papaya seed (Carica papaya L.) by observing SOD activity of blood with adenochrom method and MDA plasma level with TBARs method in mice given swimming stress. Mice was given ethanol extract of papaya seed for 7 days and swimming for 65 minutes on the 7th day. The study used 36 mice which divided into six groups, which were group I (normal), group II (negative control), group III (positive control: vitamin E), group IV (dose of 0,105 g/kg bw), group V (dose of 0,210 g/kg bw), and group VI (dose of 0,420 g/kg bw). The result showed that SOD activity of group I, II, III, IV, V, and VI was 89±6,4329 U/mL; 47,37±3,8225; 85,56±3,9210; 46,35±10,0929; 58,89±3,8271; 81,59±5,7700 U/mL, respectively, and MDA plasma level of group I, II, III, IV, V, and VI was 1,6831±0,0942; 6,3567±0,1046; 1,3697±0,2254; 4,7914±0,2139; 3,8231±0,3574 and 1,7560±0,1215 nmol/mL, respectively. This is indicated that dose of 0,420 g/kg bw of ethanol extract of papaya seed gave the highest antioxidant effect.
