Isolasi, Karakterisasi dan Uji Aktivitas Biologi Asam Kojat dari Fermentasi Cair Aspergillus Flavus LS03

Aspergillus flavus LS03 merupakan salah satu jenis jamur yang diisolasi dari tanah rhizosfer daerah Lombok Barat, yang menunjukkan potensi sebagai inhibitor α-glukosidase dan penangkap radikal bebas DPPH pada tahap awal screening aktivitas. Penelitian ini bertujuan untuk mengidentifikasi dan mengkarakterisasi metabolit sekunder sebagai antidiabetes dan antioksidan dari kapang A. flavus LS03 yang ditumbuhkan pada media cair kaldu kentang (PDB). Pengujian aktivitas antidiabetes dilakukan berdasarkan penghambatan enzim α-glukosidase sedangkan antioksidan didasarkan pada kemampuan peredaman radikal bebas DPPH. Hasil pemisahan dan pemurnian diperoleh isolat aktif (1) yang diidentifikasi sebagai 5-Hidroksi-2-(hidroksimetil)-1,4-piran (asam kojat) berdasarkan data spektroskopi. Asam kojat (1) menunjukkan potensi kuat sebagai inhibitor enzim α-glukosidase dan radikal bebas DPPH dengan nilai IC50 masing-masing sebesar 35,34 dan 149,9 µg/mL.
